Abstract:
In this study, eight populations of four
Soroseris Stebbins species were used as experimental materials. The DNA content was determined by flow cytometry based on WPB and Galbraith's buffer and ploidy was determined by traditional chromosome tableting. Results showed that, after comparing the two dissociation buffers, WPB was found to be the most suitable for
Soroseris. In addition,
S. hirsuta (Anth.) Shih had the smallest genome and the polyploidy level had little effect on the 1Cx-value in the four species. Furthermore, this study also found that seedlings formed by seed germination could be used as experimental materials instead of young leaves that are difficult to obtain.
