甘薯品种的SRAP遗传多样性分析

Genetic Diversity Analysis of Sweet Potato Based on SRAP Markers

  • 摘要: 利用分子标记SRAP技术,对36个甘薯品种进行了DNA多态性分析。选取6对引物扩增甘薯基因组DNA,共获得112条带,其中110条为多态性条带,平均每对引物提供18个标记信息。由UPMGA方法得到的聚类分析结果表明了36个品种间的遗传关系。相似系数在0.62-0.92之间,品种间有较高的遗传相似性;在聚类树中,亚洲品种和美洲品种之间没有明显的遗传分化,表明甘薯品种间没有明显的地理差异;近缘野生种与育成品种聚在一起,没有明显的遗传分化,这意味着它们之间有较高的遗传相似度;中国育成品种聚在一起,表明它们之间的遗传相似度很高。

     

    Abstract: A molecular marker SRAP(sequence-related amplified polymorphism) was used to detect DNA polymorphism among the 36 sweet potato(Ipomoea batatas)accessions from different regions.Six pairs of primers with polymorphism among 36 accessions were used to amplify the genomic DNA.A total of 112 polymorphic bands were generated,110 of them were polymorphic bands,with an average of 18 maker bands per pair of primers.The dendrogram generated by UPMGA showed the genetic relationship among 36 accessions.The genetic similarity among 36 accessions varied from 0.62 to 0.92.In the dendrogram,there was no obvious genetic diversity between Asian accessions and American accessions,indicating that there was not geographic differentiation in sweet potato.Wild relative I.trifida(6×) clustered together with other sweet potato cultivars without distinct genetic differentiation,which suggested there was high genetic coefficient between them.And there was high genetic similarity among Chinese cultivars because they clustered together.
